Investigación
La investigación del grupo de Virología Molecular está centrada en el estudio de la variación genética del VIH-1 y la evolución viral tanto por enfoques "in vitro" como "ex vivo" enfocado en las siguientes líneas:
- Pacientes no progresores. Estos pacientes mantienen el control de la enfermedad en ausencia de tratamiento antirretroviral por lo que se han propuesto como un modelo de cura funcional. Nuestro objetivo es estudiar la contribución de los factores virales en el control de la enfermedad mediante la caracterización biológica y la evolución viral en individuos con cargas virales indetectables (controladores de élite, CE), en comparación con individuos que presentan otros patrones de control viral.
- Envoltura viral: esta proteína viral es clave para determinar la eficacia viral. Por ello, su funcionalidad afecta de forma significativa a la progresión de la infección. En colaboración con Dr. Blanco y Dr. Valenzuela estudiamos qué eventos concretos (unión a CD4, fusogenicidad, etc) están relacionados con la funcionalidad de la envoltura. Para ello, hemos analizado envueltas de individuos con distintos patrones de progresión. Parte de ellas han sido cedidas al biobanco de envueltas de la Red de Investigación en SIDA para su uso.
- Doble infección: La infección por más de una variante viral (ya sea mediante co o superinfección) puede tener consecuencias en la patogénesis de la infección. En este sentido en nuestro grupo se han analizado diferentes aspectos de la DI, su detección en pacientes no progresores, su prevalencia e incidencia en España, así como la influencia de la DI sobre la respuesta neutralizante.
- Epidemiología Molecular: En el grupo de VM hemos analizado la evolución del virus a lo largo de la epidemia en España y en otros países (Holanda, Italia, Alemania, Uruguay, Panamá, Brasil, etc).
- Estudio del papel de distintos residuos aminoacídicos de la retrotranscriptasa del VIH-1 en la función enzimática y en la capacidad de replicación utilizando un clon molecular infeccioso obtenido previamente por el grupo.
- Variabilidad "in vitro". El estudio de pases seriados se ha empleado detectar los mecanismos responsables de la ganancia o pérdida de eficacia viral.
- Estudios sobre antivirales. Se ha analizado la selección de mutaciones de resistencia “in vitro” frente a distintos antivirales, así como el efecto de estas mutaciones sobre la eficacia viral, y la actividad de nuevos antivirales como los inhibidores de ATR.
Diagnóstico y Referencia Virológica en Infecciones Producidas por VIH y VLTH
El grupo de investigación ofrece y realiza actividades de diagnóstico y referencia a través de la cartera de servicios del Centro Nacional de Microbiología a todo el sistema nacional de salud.
Estos servicios incluyen:
- Diagnóstico y Referencia de la infección por el VIH (1 y 2) mediante detección de anticuerpos específicos y detección del ADN proviral mediante PCR.
- Diagnóstico y Referencia de la infección por el HTLVI/II mediante la detección de anticuerpos específicos y la detección del ADN proviral mediante PCR. Cuantificación de la carga proviral del HTLV-1 mediante PCR a tiempo real.
Laboratorio de Referencia de la Unión Europea en el ámbito de los productos sanitarios para diagnóstico in vitro Laboratorio de Referencia de la Unión Europea (EURL) en el ámbito de los productos sanitarios para diagnóstico microbiológico in vitro (IVD) de VIH y HTLV (Reglamento 2023/2713 de 5 de diciembre de 2023.) Nuestra labor es confirmar la fiabilidad y eficacia de los dispositivos para la detección de estos patógenos y garantizar sus requisitos específicos de funcionamiento mediante pruebas de laboratorio antes de que puedan ser comercializados en el mercado europeo.
