Mecanismos de adaptación de hongos patógenos al huésped: Morfogénesis en Cryptococcus neoformans
Uno de los principales mecanismos por los que los hongos son capaces de causar enfermedad en humanos es su capacidad de evadir la respuesta inmune y adaptarse a las condiciones ambientales que se encuentra en el huésped. En este sentido, una de las levaduras tiene que mayor capacidad de adaptación al huésped es Cryptococcus neoformans. Este hongo se encuentra en el ambiente, y se adquiere por inhalación, aunque el cuadro más típico es meningitis en pacientes inmunodeprimidos, principalmente VIH+. La principal característica fenotípica es la presencia de una cápsula de polisacárido que rodea a la célula que es considerado un factor de virulencia. Además, C. neoformans es capaz de aumentar el tamaño celular significativamente formando células “titanes”, que pueden alcanzar un diámetro de más de 70 micras. En el laboratorio estamos interesados en conocer cual es el papel de estos las células titanes en la virulencia de C. neoformans. Recientemente, hemos descrito medios in vitro en los que C. neoformans forma células pseudo-titanes, lo que nos ha permitido identificar nuevos factores y rutas involucradas en este proceso.
Mecanismos de acción a antifúngicos
En paralelo, llevamos una línea cuyo principal objetivo es caracterizar los mecanismos de acción de los antífúngicos. En concreto, se ha centrado el trabajo en el efecto de la Anfotericina B (AmB). Durante décadas se ha pensado que este antifúngico causa la muerte de las células tras la unión a ergosterol y formación de poros. Nuestros resultados indican que este antifúngico induce también un fuerte estrés oxidativo en la célula, el cual ocurre antes de que se pierda la integridad celular. Además hemos comprobado que el estrés oxidativo es necesario para la acción fungicida de la AmB. Estos resultados abren la puerta a diseñar nuevas estrategias que mejoren su eficiencia en pacientes.
Nuevas estrategias terapéuticas
El trabajo con AmB ha derivado en investigación orientada a mejorar las terapias antifúngicas. En particular, hemos utilizado la estrategia del reposicionamiento de fármacos “off-patent” para buscar nuevas actividades. Usando este abordaje, hemos identificado varios fármacos que aumentan la efectividad de la AmB frente a las principales levaduras patógenas, como el antibiótico eritromicina. Este abordaje nos ha permitido identificar fármacos con actividad antifúngica frente a patógenos emergentes, como Candida auris.

Licenciado en Biología por la Universidad Complutense de Madrid y Doctor en Biología por la Universidad Autónoma de Madrid. Realizó su tesis doctoral (2000) en el CSIC bajo la dirección de la Dra. Juana María Gancedo y tras una breve estancia posdoctoral en el mismo laboratorio, en 2001, se incorporó como al laboratorio del Dr. Arturo Casadevall (Albert Einstein College of Medicine, Nueva York), donde se especializó en la investigación de mecanismos de virulencia de hongos patógenos. En 2006 se incorporó al Centro Nacional de Microbiología del ISCIII gracias a un contrato “Ramón y Cajal”. Actualmente, ocupa la escala de Profesor de Investigación de los OPIs.

Nuria Trevijano Contador es Licenciada en Biología por la Universidad de Salamanca. Realizó su tesis doctoral bajo la dirección del Dr. Óscar Zaragoza Hernández en el Centro Nacional de Microbiología, la cual estuvo centrada en la importancia de la morfogénesis en la virulencia de la levadura patógena Cryptococcus neoformans. En 2017 se unió como posdoctoral al laboratorio de la Dra. Liise-anne Pirofski (Albert Einstein College of Medicine, Nueva York, USA), donde se centró en la caracterización de la interacción patógeno-huésped, y el efecto de anticuerpos sobre hongos patógenos. En 2020 se reincoporó al Centro Nacional de Microbiología bajo la supervisión del Dr. Óscar Zaragoza Hernández gracias a la concesión de un contrato posdoctoral de la Comunidad Autónoma de Madrid (programa Atracción del Talento).

Alba Torres Cano es licenciada en Biología Sanitaria por la Universidad de Alcalá de Henares (UAH), y realizó el máster "Microbiología Aplicada a la Salud Pública y Enfermedades infecciosas" de la UAH. Realizó su trabajo de fin de máster en el CNM bajo la dirección del Dr. Zaragoza en 2022, centrándose en levaduras patógenas. En ese año, se incorporó al ISCIII con un contrato predoctoral FPI bajo la dirección del Dr. Óscar Zaragoza.
